Rezumat:
The aim of this paper is to study the growth kinetics of a strain of Pseudomonas aeruginosa feeding on peptone under aerobic conditions. This is a part of a larger experimental project dealing with the optimisation of a preparation process of an immunomodulator product extracted from the harvested cells. The therapeutic effect was evaluated by protecting activity against bacterial infection on mice.
The experiments were conducted in a bottom driven 100 L Bioengineering® batch reactor. 42 L aqueous Organotech® peptone solution was charged, including an inoculum of 3 L pure culture cell suspension. Air was sparged at the bottom of reactor. The reactor was equipped with pH, temperature, dissolved oxygen, and agitation controllers. The operating parameters were 37 oC, 1.2 atm, 250 rpm, air inlet 15 L/min. The cell concentration was determined by a standard dry-weight method followed by off line optical density readings. The substrate consumption was followed by analysing the aminic nitrogen. The oxygen volumetric transfer coefficient was calculated using literature correlation for this type of reactor. kLaV was respectively 0.0202 s-1 from Sherwood number and 0.0269 s-1 from specific power input.
At the time of investigation a Tessier model applied for double substrate, the organic nitrogen source and the dissolved oxygen, represents with good approximation the experimental data. The estimated kinetic parameter are &miu m =0.8308 h-1, KO = 0.0011 g/L, KS = 0.0035 g/L.
