Rezumat:
Due to the implication of S-adenosylmethionine (SAM) and S-adenosylhomocysteine (SAH) on essential metabolic pathways of hormones, nucleic acids, several drug types, etc. and also due to the importance of the value of SAM/SAH ratio in incipient diagnosis of various pathological manifestation, the development of a proper method for quantification of these compounds is very important. We report here the development of two analytical methods high-performance liquid chromatography (HPLC) based. One is using the UV detection and the other one, the fluorescence detection. In our experimental conditions the most suitable method for SAM and SAH simultaneous determination was found to be HPLC with UV detection. Some steps on method validation were performed, namely validation in terms of sensitivity, accuracy (reproducibility, repeatability) and linearity domain. Determination of SAM and SAH using HPLC with UV detection proved a linear concentration range of 2.5× 10 -4-7.5×10-7 mol L-1 and a detection limit of 10-6 mol L-1, the reproducibility of the determination being good (the coefficient of variation (CV), CV < 2 for SAH determination and, respectively, CV < 6 for SAM determination).
