Rezumat:
Aim of the study. Development of an HPLC method for quantification of triclorfon in vitro released from acidic solution after static diffusion through cellulose membranes.
Material and method. A reversed-phase mechanism with DAD detection was used to determine the amount of toxic compound released.
Results. The proposed HPLC method uses a reverse-phase mechanism, a Michrosorb C18 (150×4.6mm) column, isocratic elution, 40°C column temperature. The mobile phase consists in 13% acetonitrile and 87% aqueous phase (pH 3.2 with H2SO4). The analyte detection is UV, at =2204 nm, with reference =280 4 nm. The calibration graphic is rectilinear between 50 and 5000 g/ml (r=0.9999, n=7 for trichlorfon).
Conclusion. This method was applied to determine the transferred trichlorfon directly from acidic samples without supplementary separation procedure. The toxic compound is relatively stable in water below pH 5.5; at higher pH, trichlorfon undergoes transformation to dichlorvos.
